Synopsis: 

Uterine fibroids (UFs; leiomyomas) are monoclonal tumors of the myometrium and represent the most important benign neoplastic threat to women’s health worldwide. With no long-term, non-invasive treatment option currently available for UFs, deeper insight concerning tumor etiology is key to the development of new and effective medical therapies. In this regard, our recent findings reveal that somatic mutations in Mediator subunit 12 (MED12) responsible for 70% of UFs disrupt Mediator-associated CDK8/19 activity leading to aberrant epigenomic change and genomic instability. We will discuss these findings and their possible therapeutic implications for the management of these clinically significant lesions.

Biography: 

Dr. Boyer received his Ph.D. in Biochemistry from SUNY at Buffalo where he studied the genetic and biochemical requirements for expression of glycolytic enzymes linked to metabolic disease. He received postdoctoral training at UCLA, culminating in his seminal discoveryof human Mediator, a conserved multi-subunit signal-processor through which regulatory information conveyed by gene-specific transcription factors is transduced to RNA polymerase II. As an independent investigator, he has leveraged his basic knowledge in the biochemistry of Mediator to understand how genetic variation in its constituent subunits elicit unit dysfunction, leading to diseases of development and aging.
